Endothelial IGFBP6 Suppresses Vascular Inflammation and Atherosclerosis

(Submitter supplied) It has previously been reported that expression levels of insulin-like growth factor binding protein 6 (IGFBP6) are reduced in unstable plaques compared to stable plaques in patients with coronary heart disease (CAD). However, the exact role and mechanism of IGFBP6 in endothelial homeostasis and inhibition of focal atherosclerotic plaque development in vivo remains unclear. Transcripome sequencing and bioinformatics analysis of IGFBP6-overexpressed human umbilical vein endothelial cells (HUVECs) suggest that pathway analysis and differentially expressed mRNA may help clarify the role and potential mechanism of IGFBP6 in the treatment of cardiovascular diseases.

Burdock miR8175 improves insulin sensitivity in DIO mice by targeting PTP1B and PTPRF [II]

(Submitter supplied) The incidence of type 2 diabetes (T2DM) induced by obesity is rapidly increasing. Although there are many drugs developed for type 2 diabetes, but the anti-diabetic effect of homology of medicine and food is also very popular with the majority of people. Burdock is one such food, but the molecular mechanism of anti-diabetic effect is unclear, limiting its further promotion. In recent years, studies have shown that plant mirnas can regulate host gene expression through dietary absorption, so plant miRNAs have become one of the main active ingredients in traditional Chinese medicine. more...

Extracellular vesicles from epicardial fat mediate long-distance heart-brain communication and promote atrial fibrillation associated cognitive impairment (EAT exosomal miRNA-seq and hippocampal mRNA-seq)

(Submitter supplied) 2 weeks of rapid atrial pacing increased the release of epicardial adipose tissue (EAT) exosomes. The red fluorescence of Ad-CD63-RFP traces the increase of EAT exosomes in the canine hippocampus. RNA Sequencing showed that the differentially expressed genes of cfa-miR-22e in EAT exosomes were significantly increased, and its target gene of IL33 was down-regulated in the hippocampus. Exosomes secreted by EAT in AF model canine penetrate the BBB, and activate hippocampal microglia through cfa-miR-22e/IL33 signaling pathway, which may be related to the increased risk of cognitive impairment in AF.

Next generation sequencing for miRNA profile of blood sample of goiter rat model treated by quemeiteng granule

(Submitter supplied) MicroRNAs play important roles in the regulatory processes of occurrence and progression of goiter. In the present study, whole transcriptome sequencing analysis was applied to investigate the expression profiles of miRNAs. After comparing blood samples of propylthiouracil (PTU)-induced goiter model blood samples and quemeiteng granule (QMTG) treated groups, 1 miRNA was found to be differentially expressed. more...

The mRNA and lncRNA profiles of verrucoushyperplasia (VH) and verrucouscarcinoma (VC) compared to SCC

(Submitter supplied) Squamous cell carcinoma (SCC) is the most common malignant tumor in the oral cavity, accounting for approximately 90% of all oral cancers. Verrucous carcinoma (VC), a rare low-grade variant of SCC, accounts for 2–12% of all oral malignancies and has a 5-year survival rate of approximately 50%. Although VC is considered a variant of SCC, it can be differentiated from SCC due to its benign histological characteristics. more...

lncRNA profile of CAFs derived EVs

(Submitter supplied) Perineural invasion (PNI) is a unique biological feature of pancreatic cancer and is a key cause of pancreatic cancer metastasis, recurrence and poor postoperative survival, but its mechanism is largely unclarified. Clinical sample analysis and endoscopic ultrasonographic elasticity scoring indicated that cancer-associated fibroblasts (CAFs) are closely related to the occurrence of PNI. Furthermore, CAF-derived extracellular vesicles played an extremely important role in PNI in a dorsal root ganglion (DRG) coculture model and sciatic nerve model. more...

The effect of high or low fat diet on the miRNA composition of exosomes derived from adipose tissue

(Submitter supplied) Exosomes are nanosized extracellular vesicles with lipid bilayer membranes and contain various contents, including lipids, miRNAs and proteins, all of which are widely involved in signaling pathways and genetic information processes. Exosomes derived from adipose tissues (AT-Exos) have been identified as a crucialmedium in the transmission of information from adipose tissue to itself and to other organs, including exosomes derived from inguinal white adipose tissue (iWAT-Exos), visceral white adipose tissue (vWAT-Exos) and brown adipose tissue (BAT-Exos). more...

Chemical manipulation of m1A mediates its detection in human tRNA

(Submitter supplied) N1-methyl adenosine (m1A) is a wide-spread RNA modification present in tRNA, rRNA and mRNA. m1A modification sites in tRNAs are evolutionary conserved and its formation on tRNA is catalyzed by methyltransferase TRMT61A and TRMT6 complex. m1A promotes translation initiation and elongation. Due to its positive charge under physiological conditions, m1A can notably modulate RNA structure. It also blocks Watson-Crick base pairing and causes mutation and truncation during reverse transcription. more...

Dynamic Changes in Extracellular Vesicle-Associated miRNAs Elicited by Abdominal Ultrasound in Peripheral Blood of Inflammatory Bowel Disease Patients

(Submitter supplied) Blood-based biomarkers that reliably indicate disease activity in the intestinal tract are an important unmet need in the management of patients with IBD. Extracellular vesicles (EVs) are cell-derived membranous microparticles, which reflect the cellular and functional state of their site of site of origin. As ultrasound waves may lead to an increased release of EVs, we hypothesized that application of ultrasound waves on inflamed intestinal tissue in IBD may amplify the release of tissue-specific EVs, which in turn can be detected in the peripheral blood.

miRNA seq mouse liver HFD, PCB126, Aroclor1260 exposure

(Submitter supplied) Exposure to high fat diet (HFD) and persistent organic pollutants including polychlorinated biphenyls (PCBs) is associated with liver injury in human populations and with non-alcoholic fatty liver disease (NAFLD) and steatohepatitis (NASH) in animal models. Exposure of HFD-fed male mice to the non-dioxin-like (NDL) PCB mixture Aroclor1260 or to dioxin-like (DL) PCB126 or to the combination caused steatohepatitis and differentially altered the liver proteome with pathways involving epigenetic regulation of gene expression. more...

High-throughput sequencing of oxidative stress for Human gastric cancer cell lines treatment with Bioactive polypeptide chelating selenium (ACBP-S-Se)

(Submitter supplied) The functional gene analyses revealed 911 upregulated genes and 1122 downregulated genes after ACBP-S-Se treatment in MKN-45 cells. From the functional enrichment analyses of oxidative stress-related genes, a total of 75 genes with significant differences were selected . From the gene interaction mapping, CDKN1A, CCNB1, TXN and MAP3K5 were extracted due to the roles of most genes with which they associate and to the significantly enriched pathways, which can be used as downstream molecules.according to the interaction of genes related to oxidative stress and lncRNAs, networks of oxidative stress genes and lncRNA interactions (lncRNA FPKM>5) were constructed via lncRNA trans and cis analysis (TRANS_ENERGY<-50). more...
